Can a Custom Layout guy switch to Physical Design(Pnr) Domain after having some experience in Custom Layout

If he's eager to learn PnR, why not? Quite a lot of people do both.

I have done full custom circuit design and P&R block at the same time many years ago.

So is there possibility that a custom guy can switch to PD domain down after couple of years working in Layout

Yes it is possible but you would have to work on your design skills. Just because you do layout doesn't mean you know how to bias up a transistor! In your free time just start working on the basics and building up circuits!
