Same thing. You can put a bootable DOS image on a USB key with the same tools.

Else you need USB drivers for your DOS version.

If the command prompt environment is your thing you should consider Linux as many distros have USB support and are much newer than 1980's DOS.

i think you need to change this settings from your PC BIOS to boot from USB flash drive instead of your SATA HDD, also you will need to set DOS ready on your USB flash drive in order to make it boot from and work on.
another solution but very old called "RAM drive" by using floppy disk drive 3.5", create a start up disk then insert it in the floppy disk drive and power it on, the OS which is DOS will transfer from the floppy to RAM and processor will continue.
