Dear,

I have a ULTRA96v2 board. Also I have a Xilinx USB II programmer. Could i program the ULTRA96v2 with Xilinx USB II programmer?

Probably yes, you might need to wire an adapter. Avnet has an USB to JTAG+UART pod dedicated to ULTRA96v2, I guess this is the better solution.

But i mean is it possible to program ULTRA96v2 without AES-ACC-U96-JTAG adapter board?

Should be possible, at least with Xilinx tools. Don't know if Avnet supplies specific tools that run only with their adapater.
