when ratio of inverter(wp:wn) is 2:1,

What is difference between inverter which has 600n:300n ratio and which has 40u:20u ratio.

and why this difference happen?

hi,
2:1 ratio is done ,in order to make sure that rise and fall time of the output are same.
if we increase the width values , the driving strength of the inverter (current) increases .
