how to determine IC(inversion coefficient) and how to bridge it with gain

how to determine Gm/Id and how to bridge it with gain

In analog IC design, I extract some parameters through simulation but not accurate,sometimes 50% error.
I feel Gm/Id method may be a better choice. but how to determine IC i should used in design.
however I don't know how to relate IC with gain.
In EE214 EECS240 , the design focus on GBW and SR .etc. not gain,
Can gain be priority?
As a newbie in analog ic design, i dont know how to estimate initial aspect ratio of mos.
Can you tell me what method you use (estimate uCox Vov and current to determine W/L or Gm/method)?
