If employing a plastic enclosure for a noise sensitive board perhaps an audio preamp or other forms of preamps and using copper tape for sheilding, does the copper tape need to be grounded to the negative side of a dc power supply powering the circuit?

The tape would cover the entirety of all walls inside plastic box. The circuit only has 12v power supply powering it coming from panel mounted banana jacks onto walls of box.

It helps to prevent the noise coming from external noise sources.

Is it ineffective without grounding?

Yes, without grounding mains hum and similar interferences won't be blocked.
