You can use ansoft designer for just patch antennas. if you wish to design patch antenna it is suitable(for just start point) But if you wish to design professional type antenna even patch or 3D structures, you need ansoft HFSS or other kinds. Maybe Your instructor...

U can not use this kind of system for inventory management. u can just use it for bargaining and returning of books. if you want to construct such and inventory management of books, you have to have a different way on UHF band(pasif) Cos its cheapest...

First of all i suppose you ll use 860-950 Mhz. Cos identifying objects from a distance.
Second your MCU should be powerfull enough to control Rf modul (command sets for tx, and parse operations for Rx from fr modul and compare operations in MCU)
for eproom i advice...
use and compare (it gives 50 Ω But teflon has no bulk conductivity!!!) As a probe u can not use it u have to use it as dielectric around probe)

As i understood u r using aperture coupling and using patch as metallized signal(ground plane) .But i suspect to use patch as ground plane. So deatails required. Or i assume that u connected ground wrong.(NO ground)
Resistor may work(as capacitor. i m not sure about patch ll work or not) on varactor reverse voltage polarization(series to varactors) But you have to calculate structure parameters(S parameters, gain etc.)

quarter patch varactors have to be reversed cos, varactors adjust capacitance with reverse dc voltage. Also shorting patch toground causes varactor malfunction cos, there ll be 0 valtage applied to varactor.
