I want to export my design from encounter to virtuoso given that I don't have the map file. So how can I transfer the layout ? and what are the difference between GDSII, DEF and OA ?

You can let encounter generate a map file for you, it will be wrong, but you can then open the file and modify it by hand. You just need the template to get started.

All the formats you mentioned are different. GDS is the de facto standard. I would suggest getting used to it.

Sir, can you give me a hint for how to edit the output map file ?

From what I understand is that map file just has each metal/via and it's corresponding layer number, am I correct ?....If I'm correct and assuming all metals/vias that I used in the layout are generated in the map file, then I will just go to my tech. LEF file and see the correct layer number and modify it in the map file, is this correct or I'm missing something ?

You are correct.
But the correct layer numbering does not come from the LEF file. It comes from those who developed the PDK/technology. There must be some documentation explaining the purpose and numbering of each layer. If your virtuoso setup is configured correctly, you can get the same information from it.
