Signal detection in real time continuous recording in MATLAB

Hello

I am recording modulated signal from mic using MATLAB. 16bit modulated signal is trasnmsitted at any instant for 400 milli sec from another PC using MATLAB using speaker. I dont know how to detect signal when recodring continuously. Then i ll stop recording. Only detection part is bottleneck. Please help
