How can one decide whether a line is an input or an output?

Hello all!
I was wondering how can one decide whether a line is an input or an output.

I want to create a ptotection interface for several DIO lines. I would need to decide the sense.

Some current comparators(current mirrors) could do the job? So I wiil be able to
detect the poalrity of current(sink/drawn) and select correspondent protection circuit?!?
