compute peak to peak ratio

i need to compute a signal's peak to peak ratio..... i have a doubt... do we calculate a signal's Peak to peak ratio in time or freq domain.... and ok say for a sine wave i can understand how to calculate the ratio.... like from postive to negative peak... but say its a signal with waveform like wer the waveform is just in the postive domain wer the other peak reaches near 0 but not to the negative peak... then how to calculate for this??? pls help me.. sorry for a very basic question... thank u
 

Also is der a way in matlab to compute peak to peak ratio ofa signal without having to do it manually... thank u
