access physical memory in DOS

Hi

Im looking for a sample C program or a small application that runs on DOS (not windows command line, but MS-DOS on USB stick) and allows the user to access to memory locations in a PC.
I want to access to the internal memory space of my PCI card design from DOS. In windows I could use the RW, but I dont want to boot windows, and i need a source code so can modify it and automate testing.
Most of the memory access methods on PC are using segmentation, but I need to directly access physical memory loacions.
