layout by drawing negative lines

I have a lot of chance to layout with negative film. It's very similar to split power plane. But draw and specify power planes are very complex. I alway draw negative line for layout. But it's very difficult for connection and open/close circuit checking.

Does anyone use this method. What software support this method? Thanks!
