If we talk about basic circuit of current mirror designed using MOS transistors. then how will be calculate the offset between the 2 transistors of basic cell ????
 

If you have mismatch (Avt) parameters from the foundry/fab you can calculate worst case offset values for adjacent transistors, depending on their size. Or you can run a Monte Carlo analysis if you have the corresponding mismatch set.
 

You can never calculate "the" offset. You can bound its likely
worst case and distribution.
