how can we measure mains voltage on single cycle(20ms)

you need just over 10mS for measuring peak AC voltage.
Like crutschow said --- peak detect or adc. But definitely use a resistive divider with appropriate safety features before you do so.
 

There are at least three different quantities that can be measured:
- rectified value
- rms value
- peak or peak-to-peak value

For each of them digital and analog methods can be imagined, also over a single cycle.
