I am assuming you already have a .bit file for your design...
So how to program that into the xc3s500e for a quick test?
In impact, right click on the xc3s500e device in the chain. (like on your second screenshot)
Do "Assign New Configuration File."
now select the bit file for your design.
You'll get a popup with something like "This device supports attached flash proms. Do you want to attach an SPI/BPI PROM?". For this example, answer "No".
When you left click on the xc3s500e device in the chain, you will see that you have now have several options, one of these being "Program". Now you can program your device to test your design.
