Port isolation

to measure the isolation port LO-to-IF and RF-to-LO in a mixer, there must not be the vco connected to the LO port ? what about if RF-LO-IF ports are differential and the instruments are single-ended ?

answer of your first question is No VCO must be connected to LO port.
you must convert diffrential to single ended by "balun" .
