microblaze vs nios
In my experience I found that for the Microblaze you can find a lot of examples and OS portings free of charge, for the NIOS all is by money, also for the uClinux portings.

If you really want to use a 4 bit microcontroller is better use the NEC uPD75X family, is very powerful and simple to use with an architecture similar to Intel family of microcontroller.
Today the use of 4 bit microcontroller is falling because the 8 bit price is very low.
For the 8051 family there are a lot of compiler in the web and a siple rtos may be the ucos is available with source code and a book (search on Elektroda forum) with a very good explanation how it work.
Re: asic vs fpga
In my experience, if you have a big production several thousands of pieces may be better the asic but for more limited quantity is better fpga.
